摘要
目的 :观察磷脂酰肌醇 3 激酶抑制剂LY2 94 0 0 2联合紫杉醇对裸鼠卵巢癌的抗肿瘤作用 ,为临床治疗中晚期卵巢癌提供实验资料 .方法 :将动物随机分为 4组 ,即正常组、卵巢癌模型组、紫杉醇组及LY2 94 0 0 2与紫杉醇联合给药组 ,用HO 891 0PM细胞皮下接种建立卵巢癌裸鼠皮下移植瘤模型 ,定期测量动物体质量及肿瘤直径 ;应用全自动血球分析仪进行血液学分析 ;荧光法测定动物肝脏丙二醛 (MDA)和超氧化物岐化酶 (SOD)含量 .结果 :LY2 94 0 0 2与紫杉醇联合应用对卵巢癌裸鼠全血WBC ,Hb的降低有促进恢复作用 (P <0 .0 5 ) ,对肝脏MDA的升高有一定的抑制作用 ,可升高因患肿瘤而降低的SOD(P <0 .0 5 ) .结论 :LY2 94 0 0
AIM: To evaluate the effects of combined therapy of Phosphatidylinositol 3 Kinase (LY294002) and Paclitaxel in athymic mice xenogeneic transplant model of ovarian cancer. METHODS: Forty athymic mice were randomly divided into 4 groups (control, positive, Paclitaxel alone, and Paclitaxel plus LY294002), and were treated respectively. Athymic mice xenogeneic transplant model was established by inoculation (sc) with HO 8910PM ovarian cancer cells. Body mass (BM) and diameter of tumor mass were measured, the peripheral blood was analyzed with auto analysis meter, and MDA and SOD of liver were determined with fluorometry. RESULTS: Paclitaxel plus LY294002 increased the amounts of WBC and HGB in the peripheral blood(P<0.05)and inhibited the MDA level of liver( P <0.05). CONCLUSION: LY294002 can enhance the effects of Paclitaxel in the treatment of ovarian cancer.
