巴曲酶对大鼠SAH后迟发性脑血管痉挛细胞凋亡的影响 被引量:4

摘要 目的 探讨巴曲酶防治蛛网膜下腔出血 (SAH)迟发性脑血管痉挛 (DCVS)及细胞凋亡的作用。方法 “枕大池二次注血法”建立大鼠SAH DCVS动物模型 ,采用TUNEL法和流式细胞仪动态观察巴曲酶对大鼠SAH DCVS的血管内皮细胞、平滑肌细胞及神经细胞凋亡的影响。结果 TUNEL检测发现巴曲酶组在SAH后 3d、7d血管内皮细胞、平滑肌细胞及神经细胞凋亡数均明显低于同期单纯注血组 ;流式细胞仪检测亦发现巴曲酶组凋亡率明显降低 ,与相对应单纯注血组比较均有明显差异 (P <0 .0 0 1)。结论 巴曲酶可以抑制SAH后的细胞凋亡 ;防治SAH DCVS及其所致的迟发性脑缺血、迟发性神经元损伤。 Objective To discuss the mechanism of Batroxobin in-theca injection to prevent and cure SAH DCVS and apoptosis. Methods Using the methods of “two-time blood injection in occipital cerebral fossa supplement”to construct dog SAH-DCVS model,utilizing TUNEL and Flow Cytometry to observe the effect of Batroxobin in-theca injection on vessel endothelial,smooth muscular cell and nerve cell apoptosis. Results The numbers of vessel endothelial,smooth muscular cell and nerve cell apoptosis of Batroxobin in-theca injection group were fewer than the same period pure transfusing group 3d and 7d after SAH by TUNEL; and the rate of apoptosis was obviously decreased under Flow Cytometry,which had obvious difference compared to pure transfusing blood group. Conclusion Batroxobin can prevent and cure SAH-DCVS and related DCVS,delayed nerve fiber damage and following cell apoptosis.
