摘要
目的 探讨 17β 雌二醇 (17β E2 )或醋酸甲羟孕酮 (MPA)对卵巢癌细胞株血管内皮细胞生长因子 (VEGF)mRNA表达的影响。方法 采用 10 -12 ,10 -11,10 -10 ,10 -9,10 -8mol/L 17β -E2 或10 -9,10 -8,10 -7,10 -6,10 -5mol/LMPA作用于卵巢癌细胞株SKOV3细胞后 ,用半定量逆转录聚合酶链反应 (RT PCR)检测SKOV3细胞VEGFmRNA水平的变化。结果 VEGF的PCR产物均以 6 0 6bp和4 74bp为主。随着 10 -12 ~ 10 -8mol/L 17β E2 浓度的增加 ,SKOV3细胞VEGFmRNA的表达呈递增趋势 ;10 -11,10 -10 ,10 -9,10 -8mol/L 17β E2 组与对照组比较 ,差异有显著性 (P <0 .0 5 )。随着 10 -9~ 10 -5mol/LMPA浓度的增加 ,SKOV3细胞VEGFmRNA的表达呈下降趋势 ,且 6 0 6bp的变化比 4 74bp明显 ,6 0 6bp可能对MPA更为敏感 ,MPA各组与对照组比较 ,差异有显著性 (P <0 .0 5 ) ;除 10 -8,10 -7mol/LMPA组间比较外 ,其余各实验组间比较 ,差异有显著性 (P <0 .0 5 )。结论 雌激素可上调SKOV3细胞的VEGFmRNA水平的表达 ;孕激素可下调SKOV3细胞VEGFmRNA水平的表达。
Objective To study the effect of 17β-estradiol (17β-E 2) and medroxyprogesterone acetate (MPA) on the expression of VEGF mRNA in epithelial ovarian carcinoma cell line in vitro. Methods Human epithelial ovarian carcinoma cell line SKOV3 cells were exposed to 17β-E 2 (10 -12~10 -8 mol/L) or MPA(10 -9~10 -5 mol/L), the expression level of VEGF mRNA was determined by RT-PCR. Results The PCR products of VEGF were mainly 606 bp and 474 bp. The expression of VEGF mRNA in the 17β-E 2-treated cells was significantly higher than in the untreated control (P<0.05), the expression level of VEGF mRNA increased with the increase in 17β-E 2 concentration. The expression of VEGF mRNA in SKOV3 cells treated with MPA (10 -9~10 -5mol/L) was significantly lower than in the untreated control (P<0.05), especially that of 606 bp. Conclusion Estrogen can up-regulate, while progesterone can down-regulate mRNA expression of VEGF. Further studies are needed to elucidate the mechanism.
