单纯疱疹病毒性角膜炎患者外周血T淋巴细胞亚群及细胞因子测定

The T lymphocytes subsets and interleukins changes in herpes simplex keratitis

目的 探讨单纯疱疹病毒性角膜炎患者外周血 T细胞亚群、自然杀伤 (natural killer,NK)细胞数目和血清白介素 (interleukin,IL) - 2、白介素 - 6 (IL- 6 )和 γ-干扰素 (interferon,IFN- γ)水平与本病发生发展关系。方法 收集初发性和复发性单纯疱疹病毒性角膜炎患者 86例 ,健康对照者 35例 ,应用碱性磷酸酶 -抗碱性磷酸酶法测定 T细胞亚群和 NK细胞数目 ,应用酶联免疫吸附试验检测血清 IL- 2、IL- 6和 IFN- γ水平。结果 86例患者中 ,初发患者 5 0例 ,复发患者 36例。与对照组比较 ,初发者 CD3、CD4、CD8和 NK细胞均值无显著性差异 ,CD4 / CD8小于对照组 ;复发组 CD3、CD4、CD8、CD4 / CD8和 NK细胞均值有显著性差异 ;两组患者比较无显著性差异。与对照组比较 ,初发组 IL- 6无显著性差异 ,IL- 2和 IFN- γ有显著变化 ;复发组 IL- 2显著下降、IL- 6和 IFN- γ显著升高。结论 初发性和复发性单纯疱疹病毒性角膜炎患者表现有 CD4 / CD8下降和 NK细胞数目下降 ,血清 IL- 2下降、IL- 6和 IFN- γ升高 。

Objective To explore the relationships of herpes simplex keratitis(HSK) and the changes of T lymphocytes subsets,natural killer (NK) cells,the serum levels of interleukin (IL)-2,IL-6 and interferon (IFN-γ).Methods 86 cases of HSK patients and 35 cases of healthy controls were collected.The APAAP method was used to detect the T lymphocyte subsets and NK cells.The ELISA assay was used to detect the serum levels of IL-2,IL-6 and IFN-γ.Results Among 86 cases of patients,50 cases were primary attack,36 cases were recurrent attack.The CD3?CD4?CD8 and NK cells in primary group were not significant changed than the control.The CD4/CD8 ratio was smaller than the control.The CD3?CD4?CD8?CD4/CD8 and NK cells were changed significantly than the control.The levels of IL-2 were decreased in both case groups.The IL-6 in primary group were not changed significantly,but the IFN-γ was changed significantly.The IL-6 and IFN-γ in recurrent group were changed significantly.There were not significant changed between the primary and recurrent group.Conclusions There were some immune changes in the HSK patients.The main changes included the CD4/CD8 ratio and NK cells decreasing,the serum levels of IL-2 decreasing,the IL-6 and IFN-γ increasing.
