含人反意N-ras cDNA顺序的假型逆转录病毒的电镜观察

摘要 本文应用透射电镜观察了经含人反意(antisense)N-ras cDNA顺序的假型逆转录病毒(pseudo-type retrovirus)转染,首次上清感染、多轮上清感染的人鼠双嗜性(amphotropic)辅助细胞PA317和小鼠单嗜性(ecotropic)辅助细胞φ2,在细胞中均见有该逆转录病毒存在。多轮上清液感染的细胞中所观察到的病毒颗粒数多于首轮感染的细胞。其结果与上清液中病毒滴度提高的结果一致。对照组细胞中未见有病毒颗粒存在。 The ecotropic helper cell φ2 and the amphotropic helper cell PA317 transfected or infected with pseudotype retrovirus containing human N-ras cDNA antisense were observed under electron microscopy. The successive rounds infected cells were observed as well. In the successive rounds infected cells, the number of the retrovirus is more than that existed in the first round. This result is identified with the increase in the titre of the retro-virus in the suspension of the infected cells. There was no virus or virus-like particle in the control groups.
