显示弹性纤维系统的一种鞣酸细胞化学染色方法

The Observation of Elastic Fiber System Stained by Modified Tannic Acid Method

弹性纤维是结缔组织中的一种重要成分。弹性纤维系统包括三种不同类型的纤维,分别称为弹性纤维、前弹性纤维及耐酸纤维。用常规透射电镜制备方法只能显示弹性纤维,而且其电子密度及反差均不够理想。如在电镜标本的制备过程中,加入鞣酸则能很好地显示弹性纤维的两种成分及三型纤维。鞣酸加入戊二醛固定液或鞣酸加入醋酸铀染色液的方法,均已有过报道,作者对常规固定的标本,在染色前用鞣酸处理,获得良好的效果,现介绍如下。

A modified Tannic Acid Stainning method was emploied for the examination of elastic fiber system under transmission electron microscope. The fine structures of oxytalan, elaunin and elastic fibers could be distin-guished clearly. Results revealed that this technique was stable in effects, simple and convenient in operation and was easily to learn. It could be used not only in the research of the elastic fiber system, but also in the ob-servations on other tissues. It should be considered as one of improved cytochemical staining methods and should be popularized in the scope of electronic microscope tissue sample processing.
