摘要
在电镜下观察了大鼠肝细胞核和鸭红细胞核的核小体、30nm纤维和放射环等染色质结构。应用电子能量损失谱成像技术(ESI)对染色质中DNA的磷元素进行了分析,在现有实验条件下,我们可以探测到一个核小体内的300个左右的磷原子,从而显示了核小体内DNA的分布。
By electron microscopy(EM)and electron spectroscopic imaging(ESI),we observed and analyzed the typ- ical structures of nucleosomes,30nm fibers and radial loops of rat hepatical and duck erythrocytic nuclear chro- matins.In our experimental conditions of specimen preparation and EM902 electron microscope,we have ob- tained structural and 300 phosphorus atom distributional information simultaneously of the nucleosome with fairly high resolution.
