HPLC法测定黄柏药材中小檗碱与黄柏碱的含量

Determination of Berberine and Phellodendrine in Cortex Phellodendri by HPLC

目的研究黄柏药材中小檗碱与黄柏碱的含量测定方法,为其质量评价提供依据。方法采用RP-HPLC法梯度洗脱,在同一色谱条件下测定不同批次黄柏药材中小檗碱与黄柏碱的含量。色谱条件:Merck-LichrospherRP-C18色谱柱(4.6×250mm,5μm),流速0.8mL/min,柱温25℃,检测波长284nm(黄柏碱)和245nm(小檗碱)。结果川黄柏中的小檗碱、黄柏碱的含量均高于关黄柏,其中小檗碱的含量川黄柏约为关黄柏的4倍,而黄柏碱则约为2～3倍。结论经过系统的方法学考察,该方法具有简便、专属、稳定、可重复的特点,可用于黄柏药材的质量评价。鉴于川黄柏与关黄柏上述两种成分含量相差较大,应分别制订含量限度。

Objective To establish a method for determination of Berberine and Phellodendrine in Cortex Phellodendri(CP).Methods Reverse-phase HPLC method was used for determination of Berberine and Phellodendrine in different batches of CP.The chromatographic conditions were:Merck-lichrospher RP-C18 column(4.6×250 mm,5 μm),flow rate being 0.8 mL/min,column temperature at 25 ℃,detection wavelength at 284nm for Phellodendrine and 245nm for Berberine.Results The content of Berberine was 4 times and that of Phellodendrine in Chuan CP 2～3 times as much as that of Guan CP.Conclusion The method is proved to be feasible for quality assessment of Cortex Phellodendri.Limited contents of Berberine and Phellodendrine should be laid out for the great difference in Chuan CP and Guan CP.
