摘要
利用ITS-RFLP技术对柑桔寄生线虫半穿刺线虫和短体线虫rDNA中的ITS区进行了研究,2个属的7个线虫种群均能扩增出相同的400bp的ITS1片段和1100bp的完整ITS区片段;分别用AluⅠ、HaeⅢ和SauAⅠ对ITS区的PCR产物进行酶切,各线虫种群ITS区扩增片段的AluⅠ、Sau3AⅠ酶切消化图谱完全一致,而HaeⅢ酶切后能将短体线虫种群与半穿刺线虫种群区分开。
ITS regions of Tylenchulus semipenetrans and Pratylenchus spp. were studied by ITS-RFLP. A 400bp fragment of ITS1 and a 1100bp fragment of whole ITS region were amplified by PCR from all of 7 populations which belong to the two generas. Then ITS region were digested by three restriction endonucleases AluⅠ, HaeⅢ and SauAⅢ. The difference between the populations of Tylenchulus semipenetrans and the populations of Pratylenchus spp. was found according to the pattern of digestion by HaeⅢ.
