摘要 用导数-比导数波谱同时测定DNA和RNA,并探讨了最佳测定条件。当DNA和RNA的浓度在最佳线性范围20-140 μg/ml内,其标准曲线方程分别为Y=0.0035X+0.0142(r=0.9982)和Y=0.0248X-0.0235(r=0.9997),检测限分别为2.0355 μg/ml和0.0359 μg/ml,回收率为87.5%-111.0%,相对标准偏差不大于6.76%。该法用于合成样品分析,结果满意。 The new method of derivative-ratio derivative spectrum for simultaneous determination of DNA and RNA is set up, and optimal detecting conditions are discussed. The standard linear equations for DNA and RNA are Y=0.0035X + 0.0142(r=0.9982) and Y=0.0248X - 0.0235(r=0.9997) respectively when their concentration falls within the optimal linear range of 20-140μg/ml. The limit of detection for DNA and RNA is 2.0355 μg/ml and 0.0359 μg/ml respectively. The recovery is 87.5%-111.0% and relative standard deviation is less than 6.76%. Its application in determination of DNA & RNA, and analysis of synthetic samples get desirable results.
