小鼠 1-细胞胚胎在含10.8,16.2,21.6和32.4 mM 乳酸钠不同 Whitten's 液中培养时,分别有2.6%,172%,8.5%和0%的胚胎发育到扩展囊胚。在4种培养液中均有60%以上的胚胎完成了第1次卵裂,发育到2-细胞期,但以后的发育则有明显差异。培养液中较高浓度乳酸钠的存在,可能是造成小鼠胚胎体外培养的“2-细胞早期阻断”的原因之一。
22.6%,17.2%,8.5% and 0% of the 1-cell mouse embryols developed intoexpanded blastocysts when cultured in modified Whitten's media containing 10.8,16.2,21.6and 32.4mM sodium lactate,respectively.The 1-cell embryos in the four different mediaunderwent the first cleavage at the same rate,and 60% of them reached 2-cell stage.Buttheir developments afterwards were different remarkably and the 10.8 mM lactate mediumworked best.The results indicated that the high lactate concentration in the culture mediummight be one of the factors responsible for the“2-cell block”of mouse embryos in vitro.
