实验性糖尿病早期大鼠背根神经节一氧化氮合酶基因表达的变化 被引量:2

摘要 目的 :观察糖尿病早期 (2周 )大鼠背根神经节 (DRG)中一氧化氮合酶 (n NOS、i NOS、e NOS)的表达变化以及胰岛素对其的影响。方法 :按 6 0 m g/ kg一次性腹腔注射链脲佐菌素 (STZ)制备胰岛素依赖性糖尿病大鼠模型(16只 ) ,造模成功后随机分成 2组 (模型组和治疗组 ) ,治疗组于造模成功后第 2天开始给予胰岛素治疗 ,正常对照组(8只 )与模型组予皮下注射同样体积的生理盐水。 2周后取各组大鼠背根神经节 ,Trizol法提取总 RNA,应用 RT-PCR法测定 3种 NOS m RNA的表达。结果 :在糖尿病早期大鼠 DRG中 ,i NOS表达明显上调 ,胰岛素能有效逆转此变化。而 n NOS和 e NOS的表达无明显变化。结论 :在早期糖尿病 DRG中 i NOS基因表达异常 。 Objective:To observe the changes of the gene expression of nitric oxide synthase (nNOS、iNOS、eNOS )in rat dorsal ganglion(DRG) of early diabetes and the effect of insulin treatment on them.Methods:Animal model of insulin-dependent diabetes mellitus were prepared by streptozotocin(STZ 60mg/kg).The models were divided into two groups (the model group and the treated group) after they were made successfully.In the second day the treated group was injected with protamine zinc insulin.At the same time,the normal group and model group were treated with the water of same cubage.Then the dorsal root ganglion of early diabetes were isolated and the RNA was extracted by using Trizol reagent and the NOS expression was detected by the method of RT-PCR.Results:The iNOS mRNA was increased in early diabetic DRG,and can be reversed by insulin treatment.while nNOS and eNOS mRNAs have no obvious change. Conclusion:The abnormal expression of iNOS mRNA in DRG of early stage of diadetes may contribute to the pathogenesis of diabetic peripheral neuropathy.
