摘要
研究了黄瓜种子经 ( 5 8± 1)℃热水老化后的活力情况 ,并通过RAPD (随机扩增多态性DNA)扩增分析其DNA损伤情况。结果表明 :热水 ( 5 8± 1)℃处理后的黄瓜种子发芽率、活力指数和活力比都随老化时间延长而明显下降 ,热水老化 3 5分钟以后 ,黄瓜种子几乎完全丧失活力 ;RAPD扩增结果表明热水老化后的黄瓜种子DNA损伤 ,老化时间越长 ,DNA损伤越严重 ,低活力种子的扩增产物谱带与对照相比 ,强带变弱或消失 ,弱带明显增多。
The paper deals with the vigor and the genomic DNA damages of cucumber seeds that were treated with hot water of(58±1℃). The results showed that the germination capacity,vigor index,and vigor ratio decreased respectively with treatment time. The vigor of cucumber seeds almost disappeared after treating 35 minutes; The results of RAPD(Random Amplified Polymorphic DNA)showed that the genomic DNA of cucumber seeds was damaged, the damages increased with treatment time, the DNA bands of seeds with lower vigor were far more than that of normal seeds.
