低密度及氧化低密度脂蛋白对内皮细胞表达血栓调节蛋白的影响 被引量:1

Effect of Low Density Lipoprotein and Oxidized Low Density Lipoprotein on the Synthesis of Thrombomodulin in Cultured Endothelial Cells

摘要 目的观察低密度及氧化低密度脂蛋白对细胞表达血栓调节蛋白的影响。方法用低密度和氧化低密度脂蛋白处理培养的ECV -304细胞 ,24h后分别收集培养液及细胞 ,用ELISA方法检测上层培养液中的可溶性血栓调节蛋白(sTM)及细胞总TM含量。结果两者均呈剂量依赖方式促进TM的表达和sTM的分泌 ,氧化低密度脂蛋白的作用明显强于低密度脂蛋白 ,而氧化低密度脂蛋白对低密度脂蛋白有氧化修饰作用。结论TM表达及sTM分泌增加可能是脂蛋白损伤细胞的结果 。 Objective To investigate the effect of low density lipoprotein(LDL)and oxidized low density lipoprotein(OX-LDL)on the expression of thrombomodulin(TM)in cultured endothelial cells and sTMin cultured supernatants.Methods Endothelial cells(ECV-304cell strain)were incubated with different concentration of LDL(50,100mg/ml),OX-LDL(50,100mg/ml)or LDL plus OX-LDL for24hours.ELISA test was carried out to analyze the cell expression of TMand sTMshedded from cell surface.Results LDL at the level of100mg/ml proˉmoted the cell expression of TM and the secretion of sTMinto the cultured medium.OX-LDL did so not only at level of100mg/ml but also at level of50mg/ml and exhibited in a dose-dependent manner.When incubated with LDL plus OX-LDL,the expression of cell and secretion of sTM from cell surface were enhanced than any of aˉgents's done respectively.Conclusions The expression of TM and secretion of sTMfrom cell surface enhanced by LDL and OX-LDL may be a consequence of cell injury,and the underlying mechanism deserved further study.
