高效液相色谱法同时测定古方生化汤中阿魏酸和川芎嗪的含量

Determination of feruic acid and tetramethylpyrazine in shenghua tang of chinese herbal combinations by HPLC

目的 :建立用反相高效液相色谱法同时测定古方生化汤中阿魏酸和川芎嗪含量的方法。方法 :以甲醇 水 (含 1%的冰醋酸 ) ,(7∶18)为流动相 ,ZorbaxC18柱 (2 5 0mm× 4 .6mm ,5 μm)为固定相 ,紫外检测波长为 2 80nm ,外标法定量。 结果 :阿魏酸和川芎嗪分离完全 ,阿魏酸在 8.0 6～ 6 4 .4 8mg·L-1的浓度范围内回归 ,方程为Y =1811.32 +1170 .33X ,r =0 .995 4 ,平均回收率为 97.9% ,RSD为 1.4 % ;川芎嗪在 4 .0 3～ 2 0 .15mg·L-1的浓度范围内 ,回归方程为Y =16 .6 0 +14 0 1.94X ,r =0 .994 1,平均回收率为 10 1.9% ,RSD为 2 .6 %。结论 :此法快捷、简便 ,准确 ,可作为生化汤的质量标准。目的 :建立用反相高效液相色谱法同时测定古方生化汤中阿魏酸和川芎嗪含量的方法。方法 :以甲醇 水 (含 1%的冰醋酸 ) ,(7∶18)为流动相 ,ZorbaxC18柱 (2 5 0mm× 4 .6mm ,5 μm)为固定相 ,紫外检测波长为 2 80nm ,外标法定量。 结果 :阿魏酸和川芎嗪分离完全 ,阿魏酸在 8.0 6～ 6 4 .4 8mg·L-1的浓度范围内回归 ,方程为Y =1811.32 +1170 .33X ,r =0 .995 4 ,平均回收率为 97.9% ,RSD为 1.4 % ;川芎嗪在 4 .0 3～ 2 0 .15mg·L-1的浓度范围内 ,回归方程为Y =16 .6 0 +14 0 1.94X ,r =0 .994 1,平均回收率为 10 1.9%

OBJECTIVE To establish a method for determining meanwhile in shenghua tang. METHODS On ZorbaxSB C 18 column (250 mm× 4.6 mm, 5 μm);with methanol-water (containing 1% acetic )=7∶18 as mobile phase and UV determination at 280 nm.RESULTS A good linearity was obtained over the range of 8.06～ 64.48 mg·L -1 (r= 0.9954) for feruic acid and 4.03～ 20.15 mg·L -1, respectively.The average recoveries of these two components were 97.9% with RSD= 1.4% (n=5) and 101.9% with RSD= 2.6% (n=5),respectively. CONCLUSION The method was simple, rapid, accurate for the simultaneous determination of both feruic acid and tetramethylpyrazine.
