摘要 目的 :探讨硒 甲基硒代半胱氨酸 (MSC)对人食管癌细胞系EC10 9细胞增殖及Bcl 2 Bax蛋白表达的影响。方法 :用荧光染色、免疫组化及免疫蛋白印迹等技术 ,在MSC的不同浓度 (0、 1、 2、 4、 8、 16 μg mL)与时间 (6、 12、 2 4、 4 8h)的作用下 ,观察其对EC10 9细胞形态的影响以及细胞核增殖抗原Ki 6 7及凋亡相关蛋白Bcl 2 Bax的表达。结果 :细胞主要表现出固缩 ,生长密度降低 ,凋亡小体形成并被吞噬等形态改变 ,未见明显细胞毒性作用。各实验组的Ki 6 7标记指数均明显低于对照组 (P <0 0 1) ,Bcl 2 Bax表达比值降低 ,并呈明显的时间和浓度依赖性。结论 :MSC有抑制EC10 9细胞系增殖及下调Bcl 2 Objective:To investigate the effect of Se-Methylselenocysteine (MSC)on Ki-67and Bcl-2/Bax protein expression in human esophageal carcinoma EC109 cell line. Methods:Using fluorescence staining,immunohistochemistry and Western blot, the expression of Ki-67 and Bcl-2/Bax protein in human esophageal carcinoma EC109 cell line were observed and analysed at different concentrations(0、1、2、4、8、16μg/mL)and times(6、12、24、48hours).Results:MSC exposure could cause cell shrinking with rugous membrane and nuclei condensed in morphology and apoptosis body appeared in cytoplasm by Acridine Orange fluorescence staining. The preceeding changes were conspicuously with the increasing dosage and no obvious cytotoxicitic changes. By immunohistochemistry and Western blot analyses, cell proliferation was inhibited inducing by MSC associated with decreased expression of Ki-67 labeling index and Bcl-2/Bax ratio following by concentration and time dependent(P<0.01).Conclusion:MSC can decrease cell proliferation and Bcl-2/Bax ratio in vitro.
