基质金属蛋白酶在视网膜缺血再灌注损伤中早期的表达及意义

摘要 目的 :观察基质金属蛋白酶 - 9(MMP— 9)及其抑制剂 - 1(TMP— 1)在大鼠视网膜缺血再灌注损伤 (RIR)早期的表达 ,探讨MMP - 9是否参与视 -血屏障 (RBR)病理损伤过程。方法 :将 38只无眼疾Wistar大白鼠随即分为两组 :正常组和缺血再灌注对照组 ,采用前房灌注加压法建立视网膜缺血再灌注模型 ,其中缺血再灌注对照组分别按 0、3、2 4h不同时间段处死动物 ,利用免疫组化法检测MMP - 9、TIMP - 1的表达 ;电镜观察BRB硝酸镧示踪。结果 :正常组视网膜未见MMP - 9及TIMP - 1表达 ,对照组再灌注后3hMMP - 9表达增高 ,2 4h呈强阳性表达且全层分布 ,TIMP - 1呈微弱表达 ,再灌注后 3h可发现硝酸镧颗粒 ,2 4h明显增多。结论 :视网膜缺血再灌注后视 -血屏障的损害与MMP - 9活性表达增高呈正相关性 (P<0 0 1) ,提示细胞外基质降解、再塑可能是视网膜缺血再灌注病理损伤的又一个重要组成部分。 Objective:To detect the early expression of Matrix Metalloproteinase-9(MMP-9) and tissure inhibitor of Metalloproteinase-1(TIMP-1) in retinal ischemic reperfusion injury in rat,and to investigate the effect of MMP-9 on pathological lesion of blood-retina barrier.Method:38 Wistar rats were randomly divided into 2 groups:Ontrol group and ischemic reperfusion group.The model of lschemic reperfusion injury was created by increasing Iop with anterior chamber perfusion and removing the perfusion 1 hour later.Rats in ischemic reperfusion group were killed at 0,3 and 24 hours after reperfusion.The expression of MMP-9 and TIMP-1 was detected by immunohis to chemistry method.La^+ was observed with electron microscope.Result:The expression of MMP-9 and TIMP-1 was negative in controls.In ischemic reperfusion group,the expression of MMP-9 increased at 3 hours after reperfusion,its higher expression was detected in all layers of retina at 24 hours.The expression of TIMP-1 was lower.La^+ was detected at 3 hours and increased significantly at 24 hours.Conclusion:The lesion of blood-retina barrier during retinal ischemic reperfusion is positively correlated with higher expression of MMP-9(P<0.01).It indicates that the degradation and reintegration of extracellular matrix (ECM) may play an important role in retinal ischemic reperfusion injury.
