单纯疱疹病毒胸苷激酶/丙氧鸟苷系统对膀胱癌体内杀伤作用的研究

摘要 目的 :探讨逆转录病毒介导的单纯疱疹病毒胸苷激酶 /丙氧鸟苷 (HSV TK/GCV)系统对膀胱癌的体内杀伤作用。方法 :用逆转录病毒载体感染鼠源性膀胱癌细胞株T739,体外观察其对GCV的敏感性。在鼠源性T739膀胱癌动物模型中 ,以逆转录病毒单次或重复感染方法介导TK基因转移 ,观察GCV治疗前后肿瘤体积大小及动物存活时间变化。结果 :TK基因导入T739后获得了GCV敏感性 ,体内肿瘤生长受到抑制 ,动物存活时间延长 ,但单次和重复感染组之间无差别。结论 :逆转录病毒成功介导膀胱癌体内基因转移 ,HSV TK/GCV系统在体内对膀胱癌有杀伤作用 ,其可望对膀胱癌作为基因治疗。 Objective:To observe the effect of retrovirus-mediated HSV-TK/GCV gene therapy on murine bladder cancer in vitro or vivo.Method:HSV-TK gene was transferred into murine bladder cancer cell line T739 by retrovirus in vitro, its sensitivity to GCV was observed. In syngeneic murine T739 bladder cancer cell line peritoneum cavity tumor model, HSV-TK gene was transferred by single or repeat injection of retrovirus. After 6 days administration of GCV, tumor volume and animal survival time were investigated.Result:In vitro T739TK cells were sensitive to GCV. Tumor growth was inhibited and animal survival time was prolonged, but no difference was seen between single or repeat injection groups.Conclusion:Conclusions: Retrovirus-mediated gene transfer in vitro or vivo on bladder cancer was effective, HSV-TK/GCV system has a killing effect on bladder cancer.
