摘要
目的 :探讨Survivin反义核酸对拓扑替康 (TPT)诱导卵巢癌细胞SKOV3凋亡的影响。方法 :在光镜下观察Survivin反义核酸与TPT联合应用后SKOV3凋亡的形态学改变 ,通过DNA电泳检测SKOV3细胞凋亡的情况 ;应用MTT法检测SKOV3细胞对TPT的敏感性。结果 :Survivin反义核酸可明显促进TPT诱导的SKOV3凋亡 ,呈现出特征性的DNA梯状带 ;TPT对SKOV3、SKOV3/neo、SKOV3/SVVas细胞的IC50 值分别为 (1 6 0 .0 0± 6 .36 ) μg/L、(1 5 8.0 0±6 .32 ) μg/L、(4 2 .0 0± 2 .31 ) μg/L ,其差异具有统计学意义 (P <0 .0 1 )。 结论 :Survivin反义核酸可明显促进TPT诱导的卵巢癌细胞SKOV3凋亡 。
Aim: To explore the effect of antisense Survivin RNA on topotecan induced apoptosis in human ovarian cancer cell line SKOV3. Methods: Changes of apoptosis induced by Survivin antisense RNA with topotecan in morphology were detected by light microscope. MTT assay were tested to investigate the sensibility of transfected cells to topotecan. Meantime, apoptosis was identified by DNA electrophoresis. Results: Survivin antisense RNA obviously enhanced the apoptosis of SKOV3 cells induced by topotecan. By MTT assay, the IC 50 of topotecan to SKOV3, SKOV3/neo,and SKOV3/SVVas cells were(160.00±6.36)μg/L, (158.00±6.32) μg/L ,and(42.00±2.31) μg/L,respectively. The difference was significant by statistic analysis ( P <0.01). DNA ladder was viewed on agarose gel. Conclusion: Survivin antisense RNA obviously enhances the apoptosis of SKOV3 cell induced by topotecan and promotes the sensitivity of chemotherapy drug to ovarian cancer cells.
