拓扑替康和更昔洛韦单独或联合应用对SKOV3/tk细胞生长的影响

摘要 目的 :探讨更昔洛韦 (GCV)与拓扑替康 (TPT)单独或联合应用对携带有hTERT启动子调控的HSV tk基因的SKOV3细胞生长的影响。方法 :将hTERT启动子调控的HSV tk基因重组逆转录病毒载体成功转染人卵巢癌SKOV3细胞 (SKOV3/tk细胞 ) ,用MTT法观察SKOV3/tk细胞对GCV、TPT的敏感性 ;观察合用GCV和TPT对SKOV3/tk细胞的杀伤作用。结果GCV及TPT对SKOV3/tk细胞的最低有效浓度分别为 1 0mg/L和 1 0 μg/L ,1 0 0mg/LGCV与TPT合用时TPT对SKOV3/tk细胞的最低有效浓度为 5 μg/L。 结论 :GCV合用TPT较TPT单用更能抑制携带有HSV Aim: To investigate the effect of ganciclovir and topotecan on SKOV3 cell transfected HSV tk under the control of hTERT promoter. Methods: The recombinant vectors carrying hTERT promoter and HSV tk gene was constructed and transfected into SKOV3 cells. The sensitivity of the transfected cells to GCV, topotecan, GCV with topotecan were observed using MTT method, respectively. Results:The growth of the transfected cells was greatly inhibited by GCV administration at 10 mg·L -1 and topotecan at 10 μg·L -1 . Topotecan dosage could be decreased to 5 μg·L -1 when combined with GCV 100 mg·L -1 .Conclusion: The growth of SKOV3 cells transfected HSV tk gene under the control of hTERT promoter could be greatly inhibited by GCV, and the GCV with topotecan combination shows high efficiency in killing the cells.
