摘要
目的 :探讨长期应用卡马西平 (CBZ)对大鼠小脑结构的影响。方法 :将SD大鼠随机分为正常组和实验组 ,每组 1 2只。①实验组 :给予CBZ溶液 (CBZ片在 4 5℃水浴中加热振摇溶解 )灌胃 ,浓度由 6 0g/L(灌胃 1 0d)增至 80g/L(灌胃 1 0d)、1 0 0g/L(灌胃 1 0d)、1 2 0g/L(灌胃 6 0d) ,每次 1ml/1 0 0g ,共灌胃 90d。②正常组 :正常喂养 ,每d蒸馏水灌胃。观察大鼠的肢体活动、共济协调能力与精神状态等。第 90d处死大鼠 ,取小脑组织 ,光镜观察小脑的病理组织学改变 ,用透射电镜观察小脑超微结构的变化 ,并进行图像分析。结果 :正常组无异常表现。实验组大鼠在灌胃期间出现呼吸急促、行走无力、拖腿、精神萎靡等表现。与正常组相比 ,实验组大鼠小脑突触数量减少 ,突触内囊泡明显减少 ,线粒体数量减少并且肿胀。结论 :长期服用卡马西平可造成大鼠小脑超微结构明显损害 ,是大鼠出现平衡、姿势、行走 (步态 )
Aim: To study the effect of long term administration of CBZ on the structure of rat's cerebellum. Methods: SD rats were randomly divided into normal group and experimental group.After CBZ was given to rats in the experimental group (concentration increased gradually from 60g/L to 80g/L,100g/L,120g/L) for 90 days,the observation was performed on changes of rat's cerebellum ultrastructure by using transmission electron microscope and analysising the picture of cerebellum ultrastructure Results: Compared with the normal group, the experimental group showed a decreased number of synapses?synaptic vesicles and mitochondria with swelling.Conclusion: CBZ could cause obvious damage to rat's cerebellum ultrastructure, resulting in dysfunction of cerebellum such as a decreased myodynamia and disorder of balance, posture, walking and fine movement of limbs.
