动员的外周血干/祖细胞中P21的表达与CD34^+细胞的相关性

摘要 目的 :检测动员的外周血干 /祖细胞中P2 1的表达及与CD34+ 细胞的相关性 ,探讨其在动员过程中的变化及意义。方法 :选取异基因外周血干细胞移植正常供者 9例和自体外周血干细胞移植者 (急性白血病患者 ) 6例 ,运用流式细胞术检测动员的外周血干 /祖细胞中CD34+ 细胞及其亚群 ,用免疫细胞化学的方法检测外周血动员前后P2 1的表达。结果 :动员的外周血干 /祖细胞中P2 1的表达明显高于动员前 ,二者间差异有统计学意义 (P <0 .0 5 ) ;动员的外周血干 /祖细胞中CD34+ 细胞比例与P2 1的表达率呈正相关 (r=0 .5 5 3)。结论 :外周血干 /祖细胞动员过程中P2 1表达增高 ,可能与其促进祖细胞增殖。 Aim:To study the expression of P21 and its correlation with CD34 +cells in mobilized peripheral blood stem/progenitor cells (PBSCs) and explore the change and significance of P21 in the mobilization of PBSCs. Methods: CD34 +cells and its subtype were measured using flow cytometric analysis and the expression of P21 were detected by immunocytochemical staining. Results: The expression of P21 in mobilized peripheral blood was significant higher (p<0.05) than that in immobilized and the relationship between the positive rate of P21 and the CD34 +cells was significantly positive ( r =0.553). Conclusion: P21 may be involved in the mobilization of peripheral blood stem/progenitor cells by promoting the proliferation of progenitor cells, regulating differentiation,and antiapoptotic activity.
