摘要
目的 研究成人胰岛分离纯化方法。方法 在传统的胰岛分离纯化方法的基础上 ,就供体选择、消化条件及方法、胰岛保护等方面作了一些改进 ,并在消化液和Ficoll分离液中加入大豆胰酶抑制剂 (STI)、牛血清白蛋白 (BSA)。结果 分离纯化后胰岛产量由原来的 (991± 14 4 )个胰岛·g-1组织提高至 (2 0 4 3± 2 4 9)个胰岛·g-1组织 (P <0 .0 1) ;纯度由 4 0 %提高到 6 0 %。结论 改进后的方法简单易行、经济实用 ,成功解决了传统方法中凝胶样物形成的难题 ;证实STI、BSA在成人胰岛分离纯化中同样具有保护作用 ;所获胰岛产量、纯度提高 ,并具良好的生物学活性。
Objective To study the method for isolation and purification of islets in adult humans. Methods The traditional method was modified in aspects of the selection of donor, the condition and technique for optimum digestion, and the preservation of isolated islets. Soybean trypsin inhibitor (STI) and bovine serum albumin (BSA) were added into the digestive and Ficoll separation solutions. Results The yield of islets was increased from (991±144) islets to (2 043±249) islets per gram of tissue. The purity of islets improved from 40% to 60%. Conclusion The modified method is feasible, convenient and economical. It resolved the development of gellike material in traditional method, proving that STI and BSA can protect the function and morphologic integrity of islets in adult humans, with higher islets yield and purity, and good biological activity.
