摘要
【目的】 验证UCP4基因在大鼠脂肪组织中的mRNA表达 ,探讨脂肪组织中UCP4基因mRNA表达水平变化与肥胖之间的关系。 【方法】 ①采用RT PCR技术获得大鼠腹膜后脂肪组织中UCP4基因的PCR产物 ,测序结果用BLAST软件进行序列同源性比对分析 ,以验证UCP4基因在大鼠脂肪组织中的表达 ;②建立高营养饮食诱导的肥胖大鼠模型 ,应用RT PCR技术检测正常与肥胖大鼠腹膜后脂肪组织中UCP4基因表达水平的差异。 【结果】 ①PCR产物测序结果的序列同源性比对分析证实UCP4基因表达于大鼠脂肪组织 ;②肥胖大鼠腹膜后脂肪组织中UCP4基因的表达水平显著低于正常大鼠 (P <0 .0 0 1)。 【结论】 UCP4基因表达于大鼠脂肪组织中 。
To identify whether UCP4 gene expresses in rat adipose tissue and to study the relationship between the mRNA expression of UCP4 gene in adipose tissue and obesity. ① After total RNA from rat retroperitoneal adipose tissue was extracted, the PCR product of UCP4 gene was obtained by RT PCR and then sequenced. To verify UCP4 gene expression in rat adipose tissue, nucleotide BLAST analysis was applied. ② The diet induced obese rats were established by feeding a high nutrition diet for 7 weeks. After the rats were killed, the total RNA from the retroperitoneal adipose tissue was extracted and the level of UCP4 gene mRNA expression was evaluated by RT PCR. ① The expression of UCP4 gene in rat adipose tissue was verified by BLAST analysis. ② The levels of UCP4 gene mRNA expression in the retroperitoneal adipose tissue of obese rats were significantly lower than those in normal rats ( P <0.001). [Conclusion] UCP4 gene expresses in rat adipose tissue and may be involved in obesity etiology and pathophysiology.
