摘要
目的 观察阿司匹林在大鼠脑缺血-再灌损伤时,不同治疗时间窗对脑的保护作用及脑ATP含量的影响。方法 线栓法制作局部脑缺血2 h再灌模型,分别于再灌0,1和3 h给予60 mg·kg-1阿司匹林,观察再灌24 h时脑损伤面积、水肿程度、死亡率及脑组织ATP含量的变化。结果 再灌0,1和3 h阿司匹林治疗均明显缩小再灌引起的脑损伤范围、降低死亡率,3组间无显著差异。3组均明显减轻再灌引起的脑水肿,但3 h组与0 h和1 h组有显著差异。再灌0 h组脑ATP含量与假手术组无显著差异,1 h和3 h组均低于0 h组,与溶媒组无显著差异。结论 再灌0-3 h给予阿司匹林对脑缺血再灌损伤均有保护作用,但再灌3 h时对脑水肿的保护作用已不明显,且只有再灌同时给药才能明显改善脑能量代谢。
OBJECTIVE: To explore the neuroprotective action and the effects on ATP levels of Aspirin treatment in focal cerebral ischemia-reperfusion rats with different treatment time-windows. METHODS: Right middle cerebral artery was occluded by inserting a thread through internal carotid artery for 2 h, and then reperfusion for 24 h. 60 mg·kg-1 dose of Aspirin was ig administered at 0, 1 or 3 h of reperfusion respectively. The brain injury area, mortality, and cerebral edema were evaluated. Adenosinl 5′-triphosphate (ATP) levels in the brain tissue of the occluded side were assayed by capillary electrophoresis. RESULTS: The area of injured brain and mortality in the three groups were dramatically reduced by 60 mg·kg-1 dose of Aspirin. No significant difference was discovered among the three groups. The cerebral edema of occluded side was also dramatically reduced, but edema was more serious in the group to which treatment was given after reperfusion 3 h than that in the other two groups. The ATP level was improved by Aspirin in the group of treatment at reperfusion onset, but no similar effect was discovered in the other two groups. CONCLUSION: The neuroprotective effects of 60 mg·kg-1 dose of Aspirin were observed in focal cerebral ischemia-reperfusion rats, treated in different time-windows. But its effect on cerebral edema was decreased when given after reperfusion 3 h. The effect of improving ATP level was only observed when Aspirin was administered at the same time of reperfusion. It was indicated that the time-point of Aspirin treatment was very important for its neuroprotective actions.
