摘要
目的 观察缬沙坦和苯那普利对肾病综合征大鼠 2 4h尿钠排泄、血浆与肾组织内皮素 (ET)、血管紧张素Ⅱ (AngⅡ )水平的影响。方法 2 4只成年SD大鼠分为阿霉素肾病组、缬沙坦组、苯那普利组及正常对照组。观察 2 8天后的 2 4h尿蛋白、尿钠排泄 ,血浆与肾组织ET、AngⅡ水平。结果 缬沙坦、苯那普利均可增加肾病大鼠尿钠排泄 ;缬沙坦可降低肾病大鼠 2 4h尿蛋白、降低ET水平 ,但不能降低AngⅡ水平 ;苯那普利可降低肾病大鼠 2 4h尿蛋白 ,降低ET、AngⅡ水平。结论 缬沙坦及苯那普利降低尿蛋白排泄的作用可能与降低ET水平 ,改善内皮细胞功能有关。
Objective To investigate the effects of valsartan and benazepril on proteinuria, excretion of urine sodium, endothelin (ET) and angiotensin Ⅱ (AngⅡ) concentrations in plasma and renal tissue in adriamycin nephrosis (AN) rats. Methods 24 adult SD rats were divided into 4 groups (n=6 each), with one group taken as normal control (group C) and the other three made AN models, which were allocated to 3 experimental groups: group AN (AN rats fed with normal saline), group B (AN rats fed with benazepril 3 mg·kg -1·d -1) and group V (AN rats fed with valsartan 30 mg·kg -1·d -1). The rats were caged for 28 d to undergo observations by the end of the experiment, which included 24-h urine sodium and urine protein, blood urea nitrogen, blood creatinin, levels of endothelin (ET) and angiotensin Ⅱ (AngⅡ) in plasma and renal tissue. Results Both valsartan and benazepril decreased the excretion of urine protein, increased urine sodium, and decreased the level of ET in plasma and renal tissue. The level of AngⅡ was also decreased by benazepril significantly, but not by valsartan. Conclusion Valsartan and benazepril can improve the endothelial function of AN rats, decrease urine protein and improve the excretion of urine sodium.
