摘要
目的 了解血管内皮生长因子 (VEGF)在急性损伤后的大鼠脊髓中表达的动态变化。方法 采用改良Allen重量打击法大鼠急性脊髓损伤模型 ,用免疫组织化学方法观察了急性损伤后大鼠脊髓VEGF表达的动态变化。结果 急性损伤 12hVEGF即可在大鼠脊髓软脊膜、脊髓内的血管内皮细胞、胶质细胞和巨噬细胞内表达 ,1d达高峰 ,并持续到 3d ,7d时开始下降。结论 急性损伤可上调VEGF在脊髓血管内皮细胞、胶质细胞和巨噬细胞内的表达 ,VEGF可能通过促进血管内皮细胞生长对损伤神经元起一定的保护作用。
Objective To investigate the time course of vascular endothelial growth factor (VEGF) expression in rats spinal cord following acute injury. Methods The spinal cord injury(SCI) model was made by modulated Allen's weight drop method. The expressions of VEGF was assayed by using immunohistochemical staining. Results At 12 h after the injury,expression was induced by acute injury in vascular endothelial cells of pial and intramedullary vessels; in astrocytes,mainly from the white matter;and in intramedullary macrophages. It peaked on the first day, and maintained 3 days,7 days after the injury the expression began to decrease. Conclusion Acute injury can induced the expressions of VEGF.VEGF may play an important role in acute spinal cord injury by the maintenance and induction of growth of vascular endothelial cells.
