摘要 目的 探讨乳腺癌中c-myc、Rb蛋白及cyclin E、cyclin D1等细胞G1期调控蛋白的表达、相互关系及其意义。方法 应用免疫组化检测57例乳腺癌患者组织中4种蛋白的表达情况。结果 c-myc、cyclin E及cyclin D1在乳腺癌中表达的阳性率均高于乳腺纤维腺瘤(p<0.05);Rb在乳腺癌中的阳性率低于乳腺纤维腺瘤(p<0.05)。乳腺浸润性导管癌中cyclin E的表达与c-myc蛋白正相关。乳腺导管内癌中cyclin D1的表达与Rb蛋白正相关。结论 在乳腺浸润性导管癌发展的不同阶段,细胞周期异常的机制可能不同。cyclin D1在乳腺导管内癌中的过表达可能部分依赖于Rb蛋白的表达。c-myc引起的细胞周期调控异常在乳腺癌发病中为较晚期事件。细胞周期调控成分的异常与乳腺癌的发病机制关系密切。 Objective To evaluate the significance of c-myc, cyclin E, cyclin D2 and Rb expression in breast carcinoma and the relationship among the four proteins. Methods Expression of c-myc, cyclin E, cyclin D1 and Rb was detected with irnmuno-histochemical method in 57 cases of human broast cancer. Results The positive rate of c-myc, cyclin E and cyclin D1 expes-sion in the breast cancer were higher than those of fibroadenoma ( P < 0.05 ); for the Rb protein, the positive rate was 57.87% in the breast cancer, but in the fibroadenoma, it was 86.67% , and there was a significant difference beteween them ( P < 0.05 ) . In the invasive ductal carcinoma, there was a positive relationship beteween the expression of cyclin Dl and Rb. Conclusion In the different phases of the invasive ductal carcinoma, the pathogenesis of cell cycle abnomality might be different. In the in-traductal cancer, the cyclin Dl overexpression may be partly dependent on the Rb expression. The abnormality of cell cycle regulation in the breast carcinoma caused by c-myc overexpression might be the later molecular event.
