辣椒胞质雄性不育恢复基因的RAPD标记 被引量:23

摘要 为定位和克隆辣椒8001恢复系中恢复基因提供基础,用9704A8001 F2代群体开展了辣椒胞质雄性不育恢复基因分子标记研究.通过RAPD-BSA分析法,找到了一个与辣椒细胞质雄性不育恢复系8001中的恢复基因紧密连锁的RAPD标记OPL09-763,测定了该标记带的序列,并进行了同源性比较.该标记带与恢复基因之间的交换值为4.17%,遗传距离为4.18 cM. With F2 segreganting population of 9704A×8001,molecular markers linked to a fertility restorer (Rf)gene for cytoplasmic male sterility in pepper(Capsicum annuum L.)were screened by bulked segregant analysis using RAPD-PCR.A RAPD fragment specific to the bulked DNA samples from male-fertile F2 plants were identified.Further linkage analysis performed with 120 individuals in the F2 segregating population showed that the RAPD marker OPL09-763 linked to the Rf gene in restorer 8001 and positioned 4.18 cM from the Rf gene.The exchange value between the marker and Rf gene was 4.17%.On the basis of sequencing,the homology was analyzed between the OPL09-763 and other homologous region in GenBank.
