花粉管通道法转基因抗虫棉外源基因的整合方式 被引量:6

Integration Patterns of Foreign Gene in the Transgenic Bt Cotton via Pollen Tube Pathway

摘要  对花粉管通道法培育的转Bt基因抗虫棉GK19、GK22、1016、1018、1022、1025、GK5、GKsu12和SGK1进行Southern杂交分析。结果表明,目的基因能够完整整合到基因组里,HindⅢ酶切位点有可能发生甲基化或丢失。除1018以外的上述8个抗虫棉的分析结果还表明,载体骨架没有整合到棉花基因组中。 Southern blot and PCR analysis showed that in the transgenic Bt cotton lines such as GK19,GK22,1016,1018,1022,1025,GK5,GKsu12 and SGK1,the integration of foreign gene in cotton was intact,and HindⅢ Restriction enzyme site was possibly lost or methylased.T-DNA backbone sequence near LB and RB board was not inserted in the chromosome of GK19,GK22,1016,1022,1025,GK5,GKsu12 and SGK1.
