摘要
将诺氟沙星(Norfloxacin)与皮肤渗透促进剂氮酮(Azone)按药剂学方法配制成透皮搽剂,应用于哺乳母猪。分别以每个乳房皮区1,2,3mL(分别含诺氟沙星100,200,300mg)的剂量涂搽于乳房区外皮,搽药后1-3h 内,分别采集乳样进行二批检测:一批以微生物法检测乳药的抑菌环,结果表明接种20μL 乳样抑菌环直径均在9-20mm 之间,大部分达11mm 以上,乳药抑菌效力相当于或超过诺氟沙星5mg/L 最低抑菌浓度(MIC)溶液的抑菌效力;另一批以紫外分光光度法检测1mL 乳样的含药量。结果表明,搽药1mL 的乳样含药量为1.20-1.90 g/L,2mL 的为1.52-2.12g/L,3mL 的为1.86-3.50g/L。2批检测结果均表明可达到透皮给药治疗相关疾病的有效剂量。本方法可通过吮吸乳药以治疗乳猪的肠型大肠杆菌病(EC),以及可用于肠道菌性乳房炎(CM)等疾病的治疗。
Norfloxacin was mixed with Azone according to pharmaceutical procedures and made up as a liniment.This preparation was appslied to the udder skins of twenty one nursing sows,another five sows was used as a control.Three dosages Which are lml,2ml,3ml representing (containing norfloxacin 100mg, 200mg,and 300mg respectively) vere used for each udder area seperately.Within 1-3 hours of application, milk samples were collected from each udder and tested for the size of inhibition zone on agar plate of pure E. Coliculture and the amount of norfloxaein that penetrated into the milk inside the udder through the skin. The results showed (1) That the tiameters of the inhibition zone ranged from 9-20 mm,most of which ex- ceeded 11 mm,equivalent to that indueed by 20 μL amounts of the norfloxaein solution in the concentration of 5 mg per L,that has been known in excess of its minimal inhibition concentration (MIC);(2)that the pene- tration rates were as follow:Applying the preparation of 1 mL,2 mL and 3 mL to the udder skin respectively, 1.20-1.90 g, 1.52-2.12 g and 1.86-3.50 g norfloxacin per L of milk were determined respectively by using spectrophtometer at 1 and 3 hours following the application.These facts indicate that the therapy of certain disease by means of transdermal penetration of such preparations can be realized.That is to say the prepara- tions made in this experiment can be used to treat not only the Enteric Colibacillosis in piglets via sucking mild from sow′s udder but also the Coliform Mastitis of dams per se.
