摘要 本文对旨在以脱毒为目的的甘蔗茎尖培养技术进行了探讨,茎尖长约2mm,成功的关键在于防止酚的污染,采用液体培养,并在培养过程中将培养物转移到新鲜的培养基中,能成功地防止酚的污染,并使茎尖成苗率达50%以上。促根率达80%以上,结果还表明,细胞分裂素是茎尖成苗所必需的激素,而萘乙酸则不是;侧茎茎尖比主茎茎尖易成苗;适当提高蔗糖的浓度及在培养基中加入活性炭均有利于茎尖苗发根,文中还对防止酚污的机理作了分析。 This paper reported the selection for the technique of sugarcane apical meristem culture which was aimed at producing virus-free plants. The key to success was about how to prevent phenol pollution. We used 2 mm apical meristems and succeeded in it by mcans of liquid culture and constantly translating materials to fresh mcdinm. Thr ratrs of plantlrt and root formation wrrr ovrr 50% and 80% respectively. Result also showed that BA was a ncccssary auxin, but NAA not; Lateral stalk was better for plantlet formation than main stalk; Increasing sucrosc concentration and adding active carbon in medinm were of benefit to the formation of roots. Bcsides, we also analyscd the mechanism of preventing phenol pollution.
