摘要 目的 :探讨 b FGF和 TGF-β对体外培养的人牙髓干细胞增殖和分化能力的影响。方法 :采用 MTT法分别测定了 b FGF和 TGF-β作用不同浓度、不同时间单独或联合作用对人牙髓干细胞增殖能力的影响。通过碱性磷酸酶活性检测、细胞形态学观察及 DSP、DMP-1免疫组化染色来检测这两种因子对人牙髓干细胞分化能力的影响。结果 :0~ 40 ng/ m L范围内 ,b FGF单独作用能显著促进人牙髓干细胞的增殖 ,且具有浓度和时间依赖性 ,而 TGF-β促增殖作用较弱 ;两者联合作用 ,促增殖作用无显著提高 ,而促分化作用显著增强 ,不仅细胞形态明显改变 ,而且碱性磷酸酶活性显著提高 ,DSP、DMP-1呈阳性表达 ,向成牙本质细胞样细胞分化。结论 :b FGF和 TGF-β对体外培养的人牙髓干细胞增殖和分化具有重要作用 ,为对探讨影响人牙髓干细胞增殖和分化的因素提供参考依据。 AIM:To investigate the effect of basic fibroblast growth factor (bFGF)and transforming growth factor-beta(TGF-β)on proliferation and differentiation of human dental pulp stem cell(hDPSC) in vitro.METHODS:MTT colorimetric method was used to detect the dose-effect and time-effect of bFGF and TGF-β on the proliferation of hDPSC.The differentiation effect was evaluated by morphological change,alkaline phosphatase(ALP) activity assay and immunohistochemistry for DSP and DMP-1.RESULTS:bFGF alone could promote the proliferation of hDPSC expressed on concentration-dependent and time-dependent,while TGF-β alone showed differentiation effect other than proliferation.Significantly greater differentiation was found in combined group comparing with individual group based on the morphological change of odontoblast,higher ALP activity and positive expression of DSP and DMP-1 .CONCLUSION:bFGF and TGF-β might play important roles on proliferation and differentiation of hDPSC.It may provide theoretic basis on further research of the proliferation and differentiateion of hDPSC.
