摘要 目的 分析旋毛虫 5 3kDaES抗原蛋白二级结构及B细胞表位。方法 应用生物信息分析软件DNASTAR和Anthep ro5 .0 ,按照Kyte -Doolittle氨基酸亲水性标准对旋毛虫 5 3kDaES抗原的亲水性基序、二级结构及B细胞表位进行分析预测。结果 在旋毛虫 5 3kDaES抗原蛋白中 ,第 1 9- 2 5、4 0 - 5 1、6 1 - 6 7、99- 1 0 5、1 1 1 - 1 1 9、1 2 6 - 1 34、1 37- 1 5 2、1 79- 1 85、1 91 - 2 0 0、2 1 9- 2 38、2 6 1 - 2 6 7、2 81 - 2 95、31 2 - 32 3、331 - 343、36 7- 372、2 90 - 393位氨基酸残基具有较强的抗原性 ,第 1 9- 2 5、4 0 - 5 1、6 1 -6 7、99- 1 0 5、1 1 1 - 1 1 9、1 2 6 - 1 34、1 37- 1 5 2、1 79- 1 85、1 91 - 2 0 0、2 1 9- 2 38、2 6 1 - 2 6 7、2 81 - 2 95、31 2 - 32 3、331 - 343、36 7- 372、390- 393位氨基酸残基可能是B细胞的表位。结论 对旋毛虫 5 Objective To predict the secondary structure and B cell epitopes of Trichinella spiralis 53kDa excretory-secretory(ES)antigen. Methods According to hydrophilicity scores of Kyte-Doolittle, hydrophilicity motifs were predicted by bio-software of DNASTAR and Anthepro5.0. Combined with the secondary structure, the B cell epitopes were predicted. Results The aa19-25?40-51?61-67?99-105?111-119?126-134?137-152?179-185?191-200?219-238?261-267?281-295?312-323?331-343?367-372和390-393 of 53kDa ES antigen of T.spiralis had the stronger antigenicity. The B cell epitope motif of 53kDa ES antigen could be located at the aa60-67,131-135,220-230.Conclusion The results will be useful to develop the new diagnostic antigen for trichinellosis.
