摘要
目的:了解自体骨骼肌卫星细胞移植治疗大鼠心肌梗死后血清血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)的改变及临床意义。方法:30只Wistar大鼠随机分为移植组(n=15)及对照组(n=15),所有大鼠经结扎冠状动脉前降支建立心肌梗死模型。将体外培养2周的大鼠自体卫星细胞以注射的方式移植到梗死区周围,4周后病理观察移植细胞的生长、增殖情况及缺血心肌毛细血管密度的改变,并测定血清VEGF浓度及血流变参数的改变。结果:卫星细胞在梗死区中可增殖分化为横纹肌纤维,与对照组相比,移植组的缺血心肌毛细血管密度犤(523.42±82.14)和(430.73±65.04)/mm2,t=2.98,P<0.01犦、血清VEGF质量浓度明显增高犤(320.05±39.29)和(271.14±37.52)μg/L,t=2.37,P<0.05犦,而某些血液流变参数明显改善犤低切全血黏度:(11.09±1.52)和(13.02±1.56)mPa·s,t=2.22,P<0.05;红细胞聚集指数:7.59±0.81和8.56±0.59,t=3.25,P<0.01;卡松黏度:(1.64±0.41)和(2.26±0.41)mPa·s,t=3.66,P<0.01;卡松屈服应力:(11.23±1.54)和(12.69±1.59)Pa,t=2.23,P<0.05犦。结论:卫星细胞在心肌梗死区中可增殖分化为横纹肌样细胞,并通过自分泌和旁分泌的形式提高血清VEGF浓度,由此改善血流变参数及缺血心肌毛细血管密度。
AIM:To investigate the changes and clinical significance of serum vascular endothelial growth factor (VEGF) of rats after the auto skeletal muscle satellite cell (SC) implantation to treat infarction myocardium. METHODS:A total of 30 Wistar rats were randomized into two groups: implantation group (n=15) and control group(n=15).Myocardial infarction models were extablished by ligating the left anterior descending artery. SCs of rats cultured for two weeks in vitro were implanted into infarction area by infection. Four weeks later, changes of the growth and proliferation of implanted cells and capillary density of ischemic myochardium were pathologically observed, and the changes of serum VEGF concentrations and hemorheological parameters were measured. RESULTS:The SCs progressively differentiated into striated muscle fibers in the myocardial infarction site, and compared with control group, the capillary density in the ischemic myocardium [(523.42±82.14)and (430.73±65.04)/mm2,t=2.98,P< 0.01], and serum VEGF mass concentration were significantly increased[(320.05±39.29) and (271.14±37.52) μg/L,t=2.37,P< 0.05].However,some hemorheological parametes improved obviously[low cut total blood coherence:(11.09 ±1.52) and (13.02 ±1.56) mPa·s,t=2.22,P< 0.05; erythrocyte aggregation index:(7.59±0.81) and (8.56±0.59),t=3.25,P< 0.01;Kasong coherence:(1.64±0.41) and (2.26±0.41) mPa·s,t=3.66,P< 0.01; Kasong yield stress: (11.23±1.54) and (12.69±1.59) CONCLUSION:The SCs can differentiate into striated muscle cells in myocardial infarction area, and can improve VEGF concentration in the form of autocrine and paracrine, so that it ameliorates the hemorheological parameters and capillary density of ischemic myocardium.Pa,t=2.23,P< 0.05].
