摘要
目的:观察压力负荷增加大鼠心肌Ⅰ和Ⅲ型胶原及其mRNA表达的改变及鹿角方对其的影响。方法:36只雄性Wistar大鼠随机分为假手术组、模型组、鹿角方组,各组12只。建立腹主动脉狭窄所致充血性心力衰竭心肌肥厚大鼠模型,观察左室质量指数(leftventricularmassindex,LVMI),采用免疫组织化学染色方法观察心肌Ⅰ,Ⅲ型胶原的改变,采用RT-PCR方法观察心肌Ⅰ型和Ⅲ型胶原mRNA的表达,并采用放射免疫分析方法检测血浆和心肌血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)水平。结果:模型组LVMI犤(3.15±0.47)/1000犦明显高于假手术组犤(2.24±0.12)/1000犦,鹿角方组LVMI犤(2.60±0.44)/1000犦与模型组比较有明显下降,差异均有显著性意义(P<0.001~0.01);模型组Ⅰ和Ⅲ型胶原均较假手术组显著升高,鹿角方组较模型组明显下降,差异均有显著性意义(P<均0.01)。模型组Ⅰ和Ⅲ型胶原mRNA的表达均较假手术组显著升高,鹿角方组较模型组明显下降差异均有显著性意义(P<均0.05)。模型组血浆和左室心肌AngⅡ水平较假手术组显著升高,鹿角方组血浆AngⅡ和左室心肌AngⅡ水平明显降低,差异均有显著性意义(P<0.001~0.01)。结论:压力负荷增加大鼠心肌Ⅰ和Ⅲ型胶原及其mRNA的表达增加;鹿角方降低压力负荷增加大鼠心肌Ⅰ和Ⅲ型胶原及其mRNA的表达;鹿?
AIM:To observe the chages of myocardial collagen type Ⅰand type Ⅲand their mRNA expression in rats with pressure overload and the effect of lujiao prescription. METHODS:A total of 36 male Wistar rats were randomized into sham operation group, model group and lujiao prescription group, 12 in each group.Model of myocardial hypertrophy caused by abdominal aortic constriction was established to observe the left ventricular mass index(LVMI). Cardiac tissue collagen type Ⅰand Ⅲ, mRNA expression of collagen type Ⅰand Ⅲ,were observed with RT PCR method.Furthermore,plasm and cardiovascular angiotensinⅡ(AngⅡ) in rats was measured with radioimmunoassay. RESULTS:LVMI of model group [(3.15±0.47)/1 000] was significantly higher than that of sham operation group[(2.24±0.12)/1 000], while that of lujiao prescription group was decreased as compared with that of model group[(2.60±0.44)/1 000],which indicated significant difference(P< 0.001-0.01);Collagen type Ⅰand Ⅲlevel of the model group was significantly higher than that of sham operation group(all P< 0.01),while that of the lujiao prescription group was remarkably decreased,which showed significant difference(all P< 0.01).Type Ⅰand Ⅲof mRNA in the model group was significantly increased as compared with sham operation group,while that of the lujiao prescription group was significantly decreased(all P< 0.05). Plasm and myocardial AngⅡlevle of the model group were significantly increased as compared with the sham operation group(P< 0.001-0.01),while those of the lujiao prescription group were remarkably decreased,which indicated significant difference(P< 0.001-0.01).CONCLUSION:Myocardial collagen type Ⅰand type Ⅲand mRNA expression in rats with pressure overload are increased.Myocardial collagen type Ⅰand type Ⅲand their mRNA expression in rats with pressure overload are decreased by lujiao prescription.Lujiao prescription for regressing myocardial fibrosis may be related to the decreasing in circulating plasm or local AngⅡlevel and expression of collagen typeⅠand type ⅢmRNA.
