白细胞介素-1β及肿瘤坏死因子-α对大鼠肝星状细胞c-fos基因表达的调节作用

Regulation of c-fos gene expression in hepatic stellate cell by IL-1β and TNF-α

目的探讨白细胞介素1β(IL1β)、肿瘤坏死因子α(TNFα)对大鼠肝星状细胞cfos基因表达的影响。方法在培养的肝星状细胞系中加入IL1β(1μg/L)、TNFα(30μg/L),于8、24、48、72h4个时间点收集细胞,提取总RNA;用逆转录定量多聚酶链反应(PCR)方法测定cfos的基因表达水平。结果IL1β组肝星状细胞cfos基因表达水平在8、24、48h3个时间点均明显高于对照组;8h最高,为对照组的4倍。TNFα组肝星状细胞cfos的基因表达水平在8、24、48、72h均明显高于对照组;8h高,为对照组的3倍。结论IL1β、TNFα可增强肝星状细胞cfos基因的表达。

Objective To investigate the influence of interleukin 1β (IL 1β) and tumor necrosis factor α (TNFα) on the gene expression of c fos in rat hepatic stellate cell.Methods Hepatic stellate cell (HSC T6) was cultured in medium containing IL 1β (1 μg/L) and TNFα (30 μg/L).The cells were collected at different incubation period (8,24,48,72 h).Total RNA of HSC was isolated and c fos gene expression levels were detected by reverse transcription polymerase chain reaction.Results C fos gene expression levels of IL 1β group were much higher than in control group after exposure to IL 1β at 8,24 and 48 h.The maximal value was 4 fold of control group 8 h.C fos gene expression levels in TNF α group were also significantly higher than those of control group at 8,24,48 and 72 h.The maximum was 3 fold of control group at 8 h.Conclusion IL 1β and TNF α might increase the gene expression of c fos in hepatic stellate cell.
