摘要
目的 :探讨大蒜新素对鼠巨细胞病毒 (MCMV )感染小鼠脾细胞Th1 /Th2类细胞因子水平的影响及其抗MCMV作用机制。方法 :复制小鼠巨细胞病毒感染模型 ,将模型小鼠随机分成大蒜新素治疗组和安慰剂组 ,于MCMV感染后 4 8h开始分别给予大蒜新素一般剂量 (2 5mg·kg- 1/d)和等量 9.0 g/L氯化钠注射液腹腔注射 ;另设大蒜新素处理组和空白对照组 ,不接种病毒 ,分别给予大蒜新素一般剂量 (2 5mg·kg- 1/d)和等量 9.0g/L氯化钠注射液腹腔注射。用ELISA法检测小鼠脾细胞Th1类细胞因子IL 2、IFNγ和Th2类细胞因子IL 4 ,IL 1 0水平。结果 :MCMV感染可导致BALB/c小鼠Th1类细胞因子IL 2表达明显下降 (P <0 .0 1 ) ,Th2类细胞因子IL 1 0表达显著增高 (P <0 .0 1 )。大蒜新素能诱导正常BALB/c小鼠和MCMV感染模型鼠脾细胞Th1类细胞因子IL 2和IFNγ的表达显著增加 (P <0 .0 1 ) ,Th2类细胞因子IL 1 0表达明显下降 (P <0 .0 1 )。结论 :大蒜新素能显著上调正常和MCMV感染模型小鼠脾细胞Th1类细胞因子IL 2和IFNγ的表达 ,并抑制Th2类细胞因子IL 1 0的表达 。
Objective:To investigate the effects of Allitridin on the expression of Th1/Th2 cytokines of spleen cells in mice infected by murine cytomegalovirus (MCMV) and its role in anti cytomegalovirus mechanisms.Method:A BALB/c mice model system of MCMV infection was established.The MCMV model group were allocated randomly into Allitridin treated group and placebo controlled group. The same dose (25 mg·kg -1 /d) and regimen of Allitridin began to be used in treated group at the 48 h after virus infection. The same volum of saline solution was injected in placebo controlled mice.The normal group were allocated randomly into Allitridin injected group and blank controlled group,25 mg·kg -1 /d of Allitridin was used. The same volum of saline solution was injected in blank controlled group . The expression levels of Th1 cytokine IL 2, IFNγ and Th2 cytokine IL 4, IL 10 in supernatant of spleen cell culture were measured by ELISA.Result:MCMV infection could markedly down regulate the expression of IL 2, significantly up regulate the expression of IL 10 of spleen cells in mice. Allitridin could induce increased expression of Th1 cytokine IL 2 and IFNγ significantly(P<0.01),decreased expression of Th2 cytokine IL 10 markedly (P<0.01).Concluion:Allitridin could increase the expression of Th1 cytokine IL 2 and IFNγ,and inhibit the expression of Th2 cytokine IL 10 in normal and MCMV infected mice, indicating a Th1 dominant state which be helpful for clearance of cytomegalovirus (CMV) in host.
