电针提高老年痴呆大鼠学习记忆能力的NO-cGMP信号通路机制 被引量:10

摘要 目的 探讨NO cGMP信号传导通路在电针治疗老年痴呆中的可能作用。方法 通过损伤穹隆 海马伞进行老年痴呆造模 ,电针“百会”、“涌泉”、“太溪”、“血海”后 ,以水迷宫、免疫组化、放免法分别进行学习记忆能力、nNOS、cGMP的检测。结果 模型组大鼠在水迷宫实验中的平均逃避潜伏期延长 ,进入盲端错误次数增多 ,与正常组比较P <0 0 1 ;电针组治疗后平均逃避潜伏期缩短 ,进入盲端错误次数减少 ,与模型组比较P<0 0 1 ;模型组海马nNOS表达和cGMP含量下降 ,电针组治疗后海马nNOS阳性神经元数目增多 ,表达增强 ,与模型组比较P <0 0 1 ;电针组治疗后海马cGMP含量与模型组比较P >0 0 5 ,但具有一定的升高趋势。结论 电针能够提高穹隆 海马伞损伤老年痴呆大鼠学习记忆能力 ,其机制与对NO cGMP信号传导通路的影响有一定关系。 Objective To investigate the effect of electro acupuncture on NO cGMP signal transduction pathway in senile dementia rats Methods The senile dementia rats were made by cutting off fimbria fornix The electro acupuncture treatment was performed on “Baihui (DU20)”, “Yongquan(KI1)”, “Taixi(KI3)” and “Xuehai(SP10)” acupoints The learning and memory ability, the expression of nNOS and the volume of cGMP were measured by water maze, immunohistochemistry and radiate immunology respectively Results The time of average escape potential was prolonged in senile dementia model group companied with the increased mistakes to enter the bind terminal of water maze compared with the control group ( P <0 01) In electro acupuncture group, the time of average escape potential was shortened,the errors were decreased compared to the model group ( P <0 01) The expression of nNOS was enhanced by electro acupuncture treatment in hippocampus compared with model group ( P <0 01) and the volume of cGMP has an increase trend to some extent ( P >0 05) Conclusions The learning & memory ability of senile dementia rats can be improved by electro acupuncture, which was correlated with NO cGMP signal transduction pathway
