阿尔茨海默病大鼠海马PKC和GDNF的变化 被引量:2

Changes of PKC and GDNF in hippocampus of rats with Alzheimer's disease

摘要 目的 选用切断大鼠穹窿海马伞作为阿尔茨海默病 (Alzheimer’sdisease ,AD)的动物模型 ,观察AD大鼠海马PKC和GDNF的变化 ,探讨其在AD发病中的作用。方法 应用放射性同位素方法检测穹窿海马伞切断AD大鼠海马PKC的活性 ,应用Westernblot方法检测穹窿海马伞切断AD大鼠海马GDNF。结果 穹窿海马伞切断AD大鼠手术侧海马PKC活性 (565 85± 1 80 70pmol·mg- 1 ·min- 1 )与正常对照组 (744 45±1 33 2 0pmol·mg- 1 ·min- 1 )比较明显降低 (P <0 0 5) ,而手术对侧 (681 50± 1 2 0 2 0pmol·mg- 1 ·min- 1 )和假手术组 (675 52± 1 35 51pmol·mg- 1 ·min- 1 )与正常对照组比较无明显差别。用Westernblot方法检测GDNF ,手术组条带浅于正常对照组和假手术组。结论 穹窿海马伞切断AD大鼠海马PKC和GDNF均降低 ,这表明在AD海马有PKC和GDNF变化 。 Objective To study the changes of Protein kinase C ( PKC) and Glial cell line derives neurotrophic factor (GDNF) in Alzheimer's disease (AD) rat models made by transection of fimbria fornix and discuss the effects of PKC and GDNF on the development of AD Methods The activity of PKC was determined by isotope and the level of GDNF was determined by Western blot Results The activity of PKC in operating side of operated group was lower than that of control group ( 565 85±180 70 pmol·mg -1 ·min -1 vs 744 45±133 20 pmol·mg -1 ·min -1 , P <0 05 ), but there were no significant changes among the side without operation of operated group (681 50±120 20 pmol·mg -1 ·min -1 ), pseudo operated group (675 52±135 51 pmol·mg -1 ·min -1 ) and normal control group The level of GDNF in operated group was lower than that of normal control group and shame operated group Conclusions PKC and GDNF, which have effects on the development of AD, were decreased in hippocampus of rats with transection of fimbria fornix
