摘要 目的 :探讨谷胱甘S肽转移酶P1(GSTP1)基因外显子 10 5位多态性与慢性阻塞性肺疾病 (COPD)易感性的关系。方法 :应用PCR RFLP技术结合聚丙烯酰胺凝胶电泳及硝酸银染色 ,对 10 4例COPD患者和 44例非COPD人群GSTP1基因外显子 10 5位多态位点进行检测 ,并根据性别、年龄及吸烟史对其易感性进行分析。结果 :( 1)COPD组与对照组GSTP1基因A/A基因型频率分别为 0 62 5、0 5 2 2 ,二者未发现差异有显著性 (P >0 0 5 ) ,两组A/G基因型频率、G/G基因型频率也未发现差异有显著性 (P >0 0 5 )。COPD组与对照组GSTP1基因 10 5位A型等位基因频率分别为 0 78、0 73 ,未发现差异有显著性 (P >0 0 5 )。 ( 2 )根据性别、不同年龄组、不同吸烟史分析 ,COPD组与对照组之间GSTP1等位基因频率的差异均无显著性 (P值均 >0 0 5 )。结论 :GSTP1基因外显子 10 Objective To investigate the correlation between the polymorphism of glutathione S-transferase P1 (GSTP1) and the susceptibility to the development of COPD. Methods The polymorphism locus at position 105 in exon 5 of GSTP1 gene from 105 patients with COPD and 44 subjects without COPD were examinated by the techniques of PCP-RFLP as well as polyacrylamide gel electrophoresis and silver stain. Their susceptibility to the development of COPD were analyzed according to their genders, ages and smoking histories. Results (1) The frequencies of GSTP1 A/A homozygous genotype in COPD group and in the control one were 0.625, 0.522, respectively and there was no significant difference between their frequencies (P>0.05). No statistical differences were found in the frequencies of GSTP1 A allele between the two groups (P>0.05). (2) The signifiant differences were not observed in the frequencies of GSTP1 A allele between the two groups according to their genders, ages and smoking histories (all P value >0.05). Conclusion Our study suggested that the polymorphism at position 105 in exon 5 of GSTP1 gene may be not related to the susceptibility to the development of COPD in our studied subjects.
