蜕皮甾酮对实验性蛛网膜下腔出血后兔脑微血管内皮细胞与中性粒细胞间黏附的影响 被引量:6

摘要 目的采用体外孵育的血性脑脊液模拟蛛网膜下腔出血(SAH)刺激因素,观察实验性SAH后兔脑微血管内皮细胞(microvessel endothelial cells, BmvECs)细胞间黏附分子-1的表达,内皮细胞与中性粒细胞间黏附变化及蜕皮甾酮的干预效应。方法 体外培养兔BmvECs,通过免疫细胞化学染色检测对照组、血性脑脊液刺激组和蜕皮甾酮治疗组BmvECs细胞间黏附分子-1表达改变,通过内皮细胞、中性粒细胞黏附模型检测BmvECs与中性粒细胞的黏附率。结果 与对照组比较,血性脑脊液刺激组BmvECs细胞间黏附分子-1免疫细胞化学染色增强,BmvECs与中性粒细胞间黏附率增加。蜕皮甾酮治疗组BmvECs细胞间黏附分子-1染色强度、BmvECs与中性粒细胞间黏附率低于血性脑脊液刺激组。结论 血性脑脊液刺激可增强内皮细胞细胞间黏附分子-1的表达和对中性粒细胞的黏附作用,蜕皮甾酮对其具有抑制作用,可能对慢性脑血管痉挛具有治疗作用。 Objective To explore the effect of ecdysterone on expression of intercellular adhesion molecule-1 (ICAM-1) and adhesion of polymorphonulear(PWN)cells to microvessel endothelial cells(BmvECs) following experimental SAH. Methods Artificial bloody cerebrospinal fluid (BCSF) was used as stimulus on BmvECs to mimic SAH status in vivo . Immunocytochemistry was used to observe ICAM-1 expression in BmvECs of control group, BCSF group and ecdysterone group. PWN-EC adhesion model was used to detect the adhesion of PWN to BmvECs.Results Following BCSF stimulation, both immunocytochemical staining of ICAM-1 and adhesion of PWN cells to BmvECs increased in BmvECs. Ecdysterone could reduce the expression of ICAM-1 in BmvECs and inhibited the adhesion of PWN to BmvECs following BCSF stimulation. Conclusions ICAM-1 expression in BmvECs and adhesion of PWN to BmvECs could be activated directly by BCSF and be reduced by ecdysterone . Ecdysterone is possible to be a valid medicine in therapy of chronic cerebral vasospasm( CVS).
