摘要
目的采用体外孵育的血性脑脊液模拟蛛网膜下腔出血(SAH)刺激因素,观察实验性SAH后兔脑微血管内皮细胞(microvessel endothelial cells, BmvECs)细胞间黏附分子-1的表达,内皮细胞与中性粒细胞间黏附变化及蜕皮甾酮的干预效应。方法 体外培养兔BmvECs,通过免疫细胞化学染色检测对照组、血性脑脊液刺激组和蜕皮甾酮治疗组BmvECs细胞间黏附分子-1表达改变,通过内皮细胞、中性粒细胞黏附模型检测BmvECs与中性粒细胞的黏附率。结果 与对照组比较,血性脑脊液刺激组BmvECs细胞间黏附分子-1免疫细胞化学染色增强,BmvECs与中性粒细胞间黏附率增加。蜕皮甾酮治疗组BmvECs细胞间黏附分子-1染色强度、BmvECs与中性粒细胞间黏附率低于血性脑脊液刺激组。结论 血性脑脊液刺激可增强内皮细胞细胞间黏附分子-1的表达和对中性粒细胞的黏附作用,蜕皮甾酮对其具有抑制作用,可能对慢性脑血管痉挛具有治疗作用。
Objective To explore the effect of ecdysterone on expression of intercellular adhesion molecule-1 (ICAM-1) and adhesion of polymorphonulear(PWN)cells to microvessel endothelial cells(BmvECs) following experimental SAH. Methods Artificial bloody cerebrospinal fluid (BCSF) was used as stimulus on BmvECs to mimic SAH status in vivo . Immunocytochemistry was used to observe ICAM-1 expression in BmvECs of control group, BCSF group and ecdysterone group. PWN-EC adhesion model was used to detect the adhesion of PWN to BmvECs.Results Following BCSF stimulation, both immunocytochemical staining of ICAM-1 and adhesion of PWN cells to BmvECs increased in BmvECs. Ecdysterone could reduce the expression of ICAM-1 in BmvECs and inhibited the adhesion of PWN to BmvECs following BCSF stimulation. Conclusions ICAM-1 expression in BmvECs and adhesion of PWN to BmvECs could be activated directly by BCSF and be reduced by ecdysterone . Ecdysterone is possible to be a valid medicine in therapy of chronic cerebral vasospasm( CVS).
