摘要
目的 观察雷公藤甲素对γ -干扰素 (IFN -γ)刺激后人眼球后成纤维细胞 (RFs)增殖和对细胞粘附分子 - 1(ICAM - 1)、白细胞抗原 (HLA) -DR表达的影响。方法 体外培养的RFs分别与IFN -γ、雷公藤甲素孵育后 ,MTT比色法检测雷公藤甲素对RFs增殖的影响 ;流式细胞仪检测ICAM - 1、HLA -DR的表达状况。结果 雷公藤甲素能以剂量依赖方式抑制RFs的增殖 ,当剂量达 10 0 μg/L时 ,RFs的增殖完全被抑制。正常人自发表达少量的ICAM - 1,不表达HLA -DR ;IFN -γ (10 0U/L)能强烈刺激ICAM - 1、HLA -DR的表达 (P <0 0 1) ;雷公藤甲素能明显抑制IFN -γ对ICAM - 1、HLA -DR的刺激作用。结论 雷公藤甲素能够抑制细胞因子介导的RFs的生物活性 。
OBJECTIVE To evaluate the effects of triptolide on cell proliferation and the expression of intercellular adhesion molecule 1(ICAM-1),human leukocyte antigen (HLA)-DR antigen induced by interferon-γ (IFN-γ) on the surface of cultured normal human retrobulbar fibroblasts in vitro METHODS After incubated with IFN-γ in the presence of triptolide for 48 hours,the proliferation of cultured retrobulbar fibroblasts (RFs) was assessed using MTT colorimetric assay,and the cell-surface expression of ICAM-1 and HLA-DR antigen were examined by flow cytometry RESULTS The proliferation of RFs was inhibited by triptolide significantly in a dose-dependent manner,and was inhibited entirely with the concentration of 100ug/L Cultured normal human RFs spontaneously and partly expressed ICAM-1,but did not express HLA-DR IFN-γ(100U/L)significantly enhanced the expression of ICAM-1 and HLA-DR on the surface of cultured normal human RFs( P <0 01),and triptolide could antagonize the inflammatory effect of IFN-γ on RFs significantly in a dose-dependent manners CONCLUSION Triptolide inhibits the activation of cultured normal human RFs in vitro induced by IFN-γ and thus may decrease the autoimmune injury to the orbit in Graves′ophthalmopathy
