淫羊藿甙对原代培养神经元缺氧缺糖损伤的保护作用 被引量:8

摘要 目的 探讨淫羊藿甙对体外原代培养神经元缺氧缺糖损伤的作用。方法 原代培养大鼠乳鼠皮质神经元,在缺氧缺糖4、6、8 h时检测正常对照组、损伤模型组、淫羊藿甙处理组的神经元细胞噻唑蓝、乳酸脱氢酶(LDH)病理学指标变化。结果 模型组在缺氧缺糖4、6、8 h后噻唑蓝值明显低于相应对照组(P<0.01),LDH漏出率明显高于对照组(P<0.05或<0.01)。淫羊藿甙组与相应模型组比较,可提高缺氧缺糖的神经元生长能力,使噻唑蓝值有所上升;LDH漏出率有所下降;改善细胞形态。 结论 淫羊藿甙对体外原代培养神经元具有保护作用,可减轻神经元缺血性损伤。淫羊藿甙有利于缺血性脑血管疾病的治疗。 Objective To investigate the effects of Icariin on anoxic and glucose-deprived injury in primary cultured neurons. Methods Primary cultured cortical neurons of Wistar rats were studied. The degree of damage to neurons of control group, model group and Icariin group (final concentration 0.25 mg/L、0.5 mg/L、1mg/L) was reflected by the cells viability, release of lactate dehydrogenase (LDH) and morphologic changes at anoxic and glucose-deprived different time points (4、6、8h), respectively. Results The absorbance of MTT in model group was significantly decreased compared with that of control group at different anoxic and glucose-deprived time points(4、6、8 h P<0.01). The release percentage of LDH of model group was significantly higher than that of control group (P<0.05 or <0.01). Compared with the model group, treatment with Icariin (final concentration 0.25 mg/L、0.5mg/L、1mg/L) in vitro could promote cell activities of anoxic and glucose-deprived injured neurons by increasing absorbance, decreasing the release of LDH and thus decreased morphologic changes of neurons. Conclusion Icariin could protect neurons against anoxic and glucose-deprived injury, which indicate that the drug might be beneficial for the treatment of ischemic brain vascular diseases.
