中国番茄黄化曲叶病毒DNAβA-Rich区的功能鉴定 被引量:4

摘要 DNAb是近年来在一些单组分双生病毒中发现的一类新型卫星DNA分子, DNAb对于这些病毒诱导典型的病害症状是必需的. DNAb上含有3个相对保守的区域: 共同区、bC1基因和A-Rich区(A含量 > 60%). 在对中国番茄黄化曲叶病毒Y10分离物(TYLCCNV-Y10)DNAb bC1基因的功能突变研究的基础上, 对TYLCCNV-Y10 DNAb上的A-Rich区进行了功能鉴定. 对TYLCCNV-Y10 DNAb A-Rich区进行缺失突变后发现, A-Rich缺失突变体依然能够在植物中稳定复制和系统移动, 因此A-Rich区对于DNAb的复制并不是必需的. 免疫捕获PCR检测发现, A-Rich缺失突变体能够包裹在TYLCCNV-Y10 DNA-A编码的外壳蛋白之中, 表明它并不能决定DNAb的包裹, 但是A-Rich区的缺失使病毒症状减弱.
