芪菖治瘫口服液对脑缺血再灌注损伤大鼠脑组织一氧化氮和一氧化氮合成酶的影响 被引量:1

Effect of qichang zhitan solution on the nitrogen monoxide and nitric oxide synthase of brain tissue in rats after ischemia-reperfusion

摘要 目的:研究芪菖治瘫口服液是否对脑缺血再灌注损伤大鼠脑组织一氧化氮和一氧化氮合成酶(nitricoxidesynthase,NOS)产生影响,为研究开发中药新药提供理论基础。方法:用三动脉夹闭法造成大鼠脑缺血模型动物,同时用芪菖治瘫口服液、补阳还五汤、维生素E灌胃给药7d,1次/d。分为手术对照组、模型对照组、芪菖治瘫口服液高剂量组、低剂量组、补阳还五汤组及维生素E组。检测大鼠脑组织一氧化氮和NOS含量。结果:模型对照组大鼠脑组织一氧化氮(17.27±2.11)μmol/g与NOS含量(0.13±0.03)μmol明显低于手术对照组犤(44.72±7.72)μmol/g(0.27±0.05)μmol,P<0.01犦;各给药组大鼠脑一氧化氮及NOS含量均不同程度低于手术对照组,而高于模型对照组,且有统计学差异。结论:芪菖治瘫口服液可明显增加脑内一氧化氮和NOS含量,改善脑组织内环境,进而保护脑组织和治疗脑梗死。 AIM:To investigate whether the qichang zhitan solution has an effect on nitrog en monoxide(NO)and nitricoxide synthase(NOS)of brain tissue in rats after ischem ia-reperfusion and to provide theoretical foundation for studying and developin g Chinese new drugs. METHODS:Three arteries of rats were blocked to establish animal models of cer ebral ischemia. At the same time,rats were fed with qichang zhitan solution,bu yang huanhu tang,vitamin E for 7 days once every day. Rats were divided into op eration group,model group,qichang zhitan high-dosage group,qichang zhitan lo w-dosage group,buyang huanhu group and vitamin E group.NO and NOS contents in brain tissue of rats were detected. RESULTS:Contents of NO and NOS of model group[(17.27±2.11) μmol/g,(0.13±0. 03) μmol/g,respectively] were obviously lower than those of operation group[(44 .72±7.72) μmol/g,(0.27±0.05) μmol,respectively](P< 0.01),and those of the drug groups were lower than those of the operation group to some extent,while hi gher than those of model group,which showed statistical difference. CONCLUSION:Qichang zhitan solution induces an obvious increase in NO and NOS i n brain tissue, and ameliorates function of brain,and thus protects brain tissue and cures cerebral infarction.
