背景:目前对Fas有关的死亡区蛋白(Fas-associateddeathdomainpro-tein,FADD)、Fas有关的磷酸酶(Fas-associatedphosphatase-1,FAP-1)研究较少,尤其它们在脑缺血再灌注中的表达情况尚不清楚。目的:探讨脑缺血再灌注后表达的时间及表达部位。设计:随机对实验动物分组。地点和材料:本研究的地点为济南军区总医院神经内二科,健康、雄性Wistar大鼠,体重320g左右。由山东省卫生防疫站实验动物中心提供。干预:用线栓法制作大鼠大脑中动脉缺血再灌注模型,用免疫组化法检测FADD,FAP-1的表达。主要观察指标:FADD,FAP-1在缺血再灌注后鼠脑中的表达。结果:脑缺血前无FADD,FAP-1的表达,缺血再灌注后即有阳性染色,缺血再灌注12h表达最多(FADD:皮层11.60±0.57,纹状体6.27±0.72,海马14.06±1.35。FAP-1:皮层6.81±1.36,纹状体4.36±0.58,海马12.35±1.21),缺血再灌注24h阳性表达最少。海马区与皮层FADD,FAP-1的表达明显多于纹状体区。结论:大鼠脑缺血再灌注引起了FADD,FAP-1的表达,FADD,FAP-1的表达可能与凋亡有关。
BACKGROUND:At present,the study on Fas-associated death domain(FADD) and Fas -associated phosphatase-1(FAP-1) is less,and particularly their expression in ischemic cerebra still remains unclear. OBJECTIVE:To explore the expressions of time and site of FADD and FAP-1 after cerebral ischemia-reperfusion. DESIGN:Randomly divide the experimental animals into different groups. SETTING and MATERIALS:The research was conducted in the Second Department of N eurology in the General Hospital of Jinan Command of PLA.Healthy male Wistar rat s weighing 320 g were provided by the Experimental Animal Center of Epidemic-pr evention Stations of Shandong Province. INTERVENTIONS:Rat model of cerebral ischemia-reperfusion was established with suture method.Immunohistochemistry was used to detect the expression of FADD an d FAP-1. MAIN OUTCOME MEASURES:Expressions of FADD and FAP-1 in cerebra of rat after i schemia-reperfusion. RESULTS:Expressions of FADD and FAP-1 were not found before cerebral ischemia ,but positive stain was found after cerebral ischemia-reperfusion(P< 0.01).The highest point of expressions was reached at 12 hours after ischemia-reperfusion [FADD:cortex(11.60±0.57),striatum(6.27±0.72),hippocampus(14.06±1.35);FAP-1:c ortex(6.81±1.36), striatum(4.36±0.58),hippocampus(12.35±1.21)],and the lowest point was reached at 24 hours after ischemia-reperfusion.The expressions of FA DD and FAP-1 in hippocampus and cortex were more than those in striatum. CONCLUSION:Cerebral ischemia-reperfusion in rats induces the expressions of F ADD and FAP-1,which may be related to apoptosis.
