摘要 用聚乙烯醇(PVA)凝胶包埋固定法对BurkholderiacepeciaJS-02细胞进行了固定化,所得凝胶具有良好的机械性和稳定性。固定化凝胶最佳质量分数为9%,最适湿细胞包埋量为0 28g/mL,固定化JS-02细胞酶活回收率为75%,连续反应6批后,固定化细胞活力为初始活力的86%。固定化细胞的最适pH为9 0,最适温度为50℃,固定化细胞的储存稳定性及操作稳定性高于游离细胞。 The whole cells of Burkholderia cepecia JS-02containing high activity D-hydantoinase were immobilized in polyvinyl alcohol(PVA) gel.The gel beads have high mechanical strength and operational stability. The optimal concentration of PVA was 8% and the optimal cell loading by direct entrapment was 0.28 g/mL. Recovery rate of D-hydantoinase activity of the immobilized cells was 75%. After 6 batches reaction, the D-hydantoinase activity decreased to 86% of the initial. The enzymatic characteristics of immobilized Burkholderia cepecia JS-02 cell were investigated. The optimum pH was 9.0 and the optimum temperature was 50 ℃. In addition, the storage stability of the immobilized cells was somewhat enhanced.
