软肝抗纤方防止四氯化碳致大鼠肝纤维化的实验研究

Experimental Studies on Ruangan Kangxian Prescription in Preventing Rats Liver Fibrosis Induced by Carbon Tetrachloride

[目的]探讨中药软肝抗纤方(由丹参、赤芍、莪术、汉防己、猪苓、枳实、柴胡、郁金、炒鳖甲、白花蛇舌草组成)预防肝纤维化的疗效和机理。[方法]复制四氯化碳(CCl4)大鼠肝纤维化模型,通过软肝抗纤方干预肝纤维化的形成,并设立阳性对照组(复方鳖甲软肝片组)、模型组及正常组,检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、透明质酸(HA)、层粘蛋白(LN)、Ⅲ型前胶原(PCⅢ)等血清学指标,并观察肝组织病理学变化及转化生长因子β1(TGF β1)免疫组化检查情况。[结果]软肝抗纤方组和复方鳖甲软肝片组均能减少模型大鼠病死率,改善肝功能和肝纤维化的血清学指标,光镜下HE染色、Masson染色和免疫组化检查均可见纤维化程度被抑制,与模型组相比有显著性差异(均P<0.05),而两给药组间差异无显著性意义(均P>0.05)。[结论]软肝抗纤方能防止CCl4致大鼠肝纤维化,药效与复方鳖甲软肝片相近,作用机制可能与拮抗TGF β1表达有关。

[Objective] To investigate the effect of Ruangan Kangxian Prescription (RKP), a prescription mainly composed of Radix Salviae Miltiorrhizae, Radix Paeoniae Rubra, Catechu, Radix Stephaniae Tetrandrae and Polyporus, for rats liver fibrosis induced by carbon tetrachloride (CC14) and to explore its mechanism. [Methods] Rats with liver fibrosis were induced by CC14 and were randomized to RKP group, normal group, Biejia Ruangan Tablet (BRT) group and model group. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), hyaluronic acid (HA), laminin (LN) , procollagen Ⅲ (PCⅢ ) were detected; hepatic histopathology and immunohistochemical result of transforming growth factor β1 (TGF β1 ) were also examined. [ Results ] Death rate of rats was reduced; serological parameters of hepatic function and liver fibrosis were improved and liver fibrosis showed by immunohistochemical method, HE staining and Masson staining was inhibited in RKP and BRT groups (P < 0.05 compared with model group). The difference between RKP group and BRT group was insignificant ( P > 0.05). [ Conclusion ] RKP and BRT have the similar effect in preventing CC14-induced liver fibrosis in rats and its mechanism may be related to the counteraction of expression of TGF β1 .
