摘要 目的 观察D-半乳糖制造老年性聋模型改进的有效性。方法 健康豚鼠40只,随机分为4组,即对照组和三个不同剂量D-半乳糖(150、200、300mg/kg)造模的老年性聋模型组。观察各组动物ABR改变,肝组织超氧化物歧化酶(SOD)活性、耳蜗及蜗核冰冻切片螺旋神经节细胞和蜗核神经细胞改变及脂褐素沉淀情况。结果 各模型组ABR和Ⅰ、Ⅲ波潜伏期显著延长,仅300mg/kg组阈值升高,肝组织SOD活性不同程度下降。模型组螺旋神经节细胞与蜗核神经细胞稀少、肿胀,尼氏小体减少,内皮细胞增生,螺旋神经节细胞核旁可见大量脂褐素颗粒。结论 D-半乳糖造模改进法是制造老年性聋动物模型的有效方法。 Objective To investigate the feasibility of a modified modeling nethod for presbycusis by the of D-galactose. Methods Forty guinea pigs were randomly divided into 4 groups,i.e. one controlling group(CG) and three modeling groups(MG1-3)with different dosages of D-galoctose(150mg,200mg,300mg/kg) to prepare the presbycusis model. Then,observed were the changes of ABR, the activity of SOD in the hepatic tissues, morphologicalchanges and lipofuscin deposition in spiral neurons and cochlear nucleus by frozen section to the cochlea and cochlear nucleus. Results Compared with that of animals in CG, Waves Ⅰ and Ⅲ of ABR were seen with significantly delayed latencies among the animals of MG1-3 and elevated thresholds in MG3, accompanied with decreased SOD activity in the hepatic tissues. In addition, significant morphological changes could also be seen in MG3,such as scarcely distributed cells of spiral ganglion and cochlea nucleus, swollen bodies of neurons, reduced number of Niss's bodies, hyperplasia of endothelia and large quantity of lipofuscin granule deposition around the nucleus of spiral ganglion cells. Conclusion The modified procedure reported here for the model preparation of presbycusis by D-galactose is an effective one to prepare this kind of model.
